The plasmid was transfected into 293H adherent cells with Sinofection reagent (Cat# STF01). After 48 h, Immunofluorescence staining of cells. Cells were fixed with 4% PFA, permeabilzed with 0.3% Triton X-100 in PBS, blocked with 10% serum, and incubated with Mouse anti-Flag Tag monoclonal antibody (CST#8146S) at 37℃ 1 hour. Then cells were stained with Goat Anti-mouse IgG secondary antibody. The fluorescent signal is detected by fluorescence microscope. Each expression experiment has negative control.

PPARGC1A/PGC1 Background Information

Peroxisome proliferator-activated receptor γ coactivator 1α (PGC1α) encoded by the PPARGC1A gene is a vital regulator of glucose and fatty acid oxidation, mitochondrial biogenesis, and skeletal muscle fiber conversion. PGC-1α is a transcriptional co-activator that has been implicated in the pathogenesis of neurodegenerative disorders. As a central regulator of mitochondrial biogenesis and metabolism, PPARGC1A acts as a powerful coactivator of many transcriptional factors that relate to steroidogenesis. The mitochondrial regulator PPARGC1A regulates mitochondrial biogenesis, reducing senescence of vascular smooth muscle cells (VSMCs). PPARGC1A upregulates autophagy reducing senescence by an SQSTM1-dependent mechanism. Evaluation in human normal lung and metastatic lung cancer cell lines revealed that the expression of PPARGC1A was upregulated in metastatic lung cancer cell lines.
